Description
Palmitoyl-CoA is a coenzyme A derivative of palmitate formed by acyl-CoA synthase. In contrast to medium- and short-chain acyl-CoA, palmitoyl-CoA cannot freely diffuse into the mitochondrial matrix. Formation of palmitoylcarnitine by CPTI is necessary prior to transfer into mitochondria for further fatty acid oxidation (ฮฒ-oxidation). To study Fatty acid oxidation using Palmitoyl-CoA, Carnitine and low amount of malate is needed on mitochondrial preparations.
Application in HRR
- Pa-CoA: Palmitoyl-CoA (Palmitoyl-Coenzyme A lithium salt; C37H65N7O17P3S ยท xLi+), Sigma-Aldrich: P9716, store at -20 ยฐC, CAS: 188174-64-3, M = 1004.94 gยทmol-1
- Preparation of 10 mM stock solution (dissolved in H2O):
- Weigh 5 mg Palmitoyl-CoA.
- Add 497.5 ยตL H2O.
- Divide into 100 ยตL portions.
- Store at -20ยฐC.
- Preparation of 10 mM stock solution (dissolved in H2O):

- Titration volume: 8 ยตL 10 mM using a 25 ยตL Hamilton syringe (2 mL O2k-Chamber).
- Final concentration: 40 ยตM.
- Comments: Please consider that the final concentration needed in the O2k-Chamber may vary according to your cell type and overtitration of Palmitoyl-CoA might inhibit the respiration.
