Application in high-resolution respirometry
Safranin is a potent inhibitor of Complex I linked respiration and of the phosphorylation system. At a concentration of 2 Β΅M, safranin can be applied for simultaneous monitoring of oxygen consumption and mtMP of mt-preparations in a CII linked substrate state.[1] Oroboros Instruments uses Safranin O from Sigma (#S2255, 25 g).
See Discussion.
Data acquisition and analysis
A step-by-step protocol of our current way of acquiring and analyzing Safranin data is given in MiPNet19.19
