Bioblast alert - in preparation
Bioblast alert 2011(03): 2011-11-24
- O2k-Publications: Fluorometry combined with high-resolution respirometry - Light guide inserted into the chamber through a black PEEK stopper[1], or optical probe operated through the O2k-glass window[2].
- β Anderson EJ, Rodriguez E, Anderson CA, Thayne K, Chitwood WR, Kypson AP (2011) Increased propensity for cell death in diabetic human heart is mediated by mitochondrial-dependent pathways. Am. J. Physiol. Heart Circ. Physiol. 300: H118-124.
- β Hickey AJ, Renshaw GM, Speers-Roesch B, Richards JG, Wang Y, Farrell AP, Brauner CJ (2011) A radical approach to beating hypoxia: depressed free radical release from heart fibres of the hypoxia-tolerant epaulette shark (Hemiscyllum ocellatum). J. Comp. Physiol. B. [Epub ahead of print]
- High-resolution terminology - term of the week: Fluorometry versus fluorimetry, >>>new List of fluorimetry terms.
- MiPNet Laboratory of the week - Auckland, New Zealand: Anthony J Hickey: (edit text: 6 O2k, guest lecturer at IOC66, joint project (Marsden), guest tutor at IOC59).
Bioblast alert #### to select
- O2k-Publication: Heart failure and mitochondrial dysfunction. Increased OXPHOS capacity after cold preservation of the human heart β a paradox resolved by a dyscoupling mechanism.
- Scientific service: Tissue storage on ice is possible for limited periods of time (several hours) with preservation of mitochondrial respiratory function, as evaluated with SUIT protocols.
- Term of the week: Uncoupled, non-coupled, dyscoupled - a hallmark of high-resolution respirometry is a matching high-resolution terminology.
- Evaluate an example of low-resolution terminology: Uncoupling was studied in stressed cells by measurement of coupled respiration, evaluation of uncoupled respiration after inhibition with oligomycin, and FCCP titration to quantify uncoupled respiration.
Bioblast alert ## to select
- O2k-Publication: Protocols for HRR: from intact cells to permeabilized muscle fibres
Bioblast alert ### to select
- O2k-Publication: Efficiency, basal respiration, and oxygen kinetics in human muscle or Mitochondrial efficiency and P/O flux ratios: new perspectives in mitochondrial physiology (see Journal club);
- Larsen FJ, Schiffer TA, Borniquel S, Sahlin K, Ekblom B, Lundberg JO, Weitzberg E (2011) Dietary inorganic nitrate improves mitochondrial efficiency in humans. Cell Metab. 13: 149-159.
- Larsen FJ, Schiffer TA, Sahlin K, Ekblom B, Weitzberg E, Lundberg JO (2011) Mitochondrial oxygen affinity predicts basal metabolic rate in humans. FASEB J. 25: 2843-2852.
- Before alert, ask to join as MiPNet Reference Lab, and present new MiPNet Reference Lab
- Term: efficiency versus P/O ratio
Bioblast alert - concept
Here is a brief summary on the concept of Bioblast alert.
Some of the following topics (not more than four per alert) will be selected for a Bioblast alert.
MiPNet O2k-Publications
- Topic: O2-kinetics; coupling; pathology; ...
- Preparation: Homogenate/isolated mitochondria; intact/permeabilized cells; permeabilized fibre/in vivo
- Protocols
MiPNet Laboratories of the World
Contact the MiPNet laboratories: show link on the wiki, ask for updates on group, references, website, perhaps major projects (HypoxiaNet), perhaps add pictures, perhaps keywords/short description. Circular: link to presented lab / special occasion (e.g. publication; conference presentation; new member; PhD thesis, ..
High-resolution terminology
- high-resolution terminology - matching measurement at high-resolution. Term of the week: Basal respiration
- Define β Measure: terminology and experimental approach
- Define β Clarify: consider discrepancies, resolve problems
MiPevents
- Conferences and Workshops
Bioblast
- What is new or updated on the website: Lists (chemicals, protocols, scientific service, ..)
- Projects in mitochondrial science (COST HypoxiaNet; Framework programs; ..)
- Links on the Bioblast website; networking in mitochondrial science, presentation of: MIG List, Mitochondrion journal and website; Mitochondrial Societies
- MitoCom News
Bioblast editors
An editor may select an O2k-Publication (or a small number of related publications), particularly from a MiPNet Reference Laboratory, may get in contact with the particular reference lab, update info on the lab, and βpresentβ the lab in the alert. A term may be selected (perhaps related to the publication), updated, discussed in our group, and then presented in the alert. An editor does not even have to present an entire alert, but may prepare individual βpointsβ, which then can be added to a particular alert. As Bioblast is updated, the updated βarticleβ (term, publication etc) can be added to the prepared points for a future alert.
