Dear O2k-Workshop participants,

1. Workshop materials

For those who have one (or more) O2k-instruments in the lab, I recommend to bring along the O2k-Compendium (O2k-Manual). During the Workshop, we will repeatedly refer to the O2k-Manual (Open access).


The fully updated O2k-Manual, O2k-Protocols, and O2k-References (pdf reprints) will be available on your personal OROBOROS-USB included in the workshop material. Bring your laptop or use our Notebooks during the workshop, making yourself familiar with the O2k-FileFinder while our tutors can provide immediate guidance. You can download the DatLab software and many DatLab demo files from the OROBOROS-USB, such that you can practice the use of DatLab. Although our software is known as user-friendly, DatLab is always a key part of a hands-on workshop. A comprehensive methodological overview on high-resolution respirometry (Pesta and Gnaiger 2012), is recommended for reading as a good preparation for the workshop. In addition to the literature, a random or targeted walk through the O2k-Catalogue provides lots of information on the various parts of the O2k and its modular extensions.

We keep the programme flexible, and all theoretical parts are discussion-oriented and interactive. In addition to the practical sessions, we will cover some fundamentals of mitochondrial physiology and protocol design. If you want to refresh your memory on a few conventional bioenergetic terms, take a look at the Bioblast Glossary on respiratory states - we will take some time to discuss the rationale for an extension of bioenergetics to mitochondrial physiology.

Important transport and organizational details

Meeting point in Bregenz for joint transfer

  1. The meeting time at Bregenz main railway station (“Hauptbahnhof”) is 2012 December 5th 11, 3 pm - (do NOT exit at “Bregenz Hafen”).
  2. The meeting point is the main parking lot (bus terminal), where our bus “IOC72” will be waiting and depart at 3 pm.

Hotel Körbersee

  1. There is no road access to Hotel Koerbersee - we are in the middle of nature.
  2. Your luggage will be transported to the hotel by cable car from the village of Schröcken. http://www.koerbersee.at/anreise.html
  3. Walk to the hotel: After unloading your luggage, the bus driver will bring you to the meeting point “Körberkreuz”. We will walk with you to the hotel (approx. 45 min walk in a wonderful scenic environment; warm and waterproof boots are strongly recommended; see picture below). Alternatively, you will be taken to the hotel by a snow cat (depending on snow conditions).

Weather, gear, activities

  1. Schröcken is an alpine village at 1600 m to 2050 m altitude. We highly recommend to bring warm clothes (warm and waterproof hiking boots, a warm jacket, gloves, hat and warm socks).
  2. As a special event, we offer an easy (snowshoe) walk and a guided tour through the “Alpmuseum” (cheese farm museum): www.alpmuseum.at. We have to ask for a small fee for the tour (see programme).
  3. Lunch breaks can be used for skiing, walks & talks and relaxing in the wellness area.
  4. !!! There is no ATM close to the Hotel, the last possibility to withdraw money is in Schröcken village centre. Please make sure to bring some cash EUROs with you (drinks, skiing, etc).

Introductory Workshop in Parallel Sessions: O2k Basic and TPP Basic

O2k Basic includes a basic introduction to the OROBOROS Oxygraph-2k, overview presentations on high-resolution respirometry (HRR), and applications in mitochondrial physiology.

Specific emphasis is placed on hands-on practical sessions with the instrument and the software DatLab, to introduce the participants into the technical and operational details of the method. The workshop programme is complemented by combined lectures (general topics; substrate-uncoupler-inhibitor titration SUIT protocols) and experiments run in "Special interest groups". Short presentations of 'Hot topics in Mitochondrial Physiology' provide participants with the opportunity to present and discuss their work, including projects in progress, methodological details and conceptual aspects.
